KẾT LUẬN
1. Xác định các gen AGPS, AGPL và SSIV là những gen góp phần quyết
định hàm lượng tinh bột của củ sắn thông qua các phương pháp real time RTPCR và phương pháp tin sinh học.
2. Đã thiết kế được 2 vector chuyển gen ở thực vật là: pK7WG2D-35S:
SSIV:T35S mang gen mã hóa SSIV tăng cường sinh tổng hợp tinh bột dưới sự
điều khiển của promoter cố định 35S, pBI121-C54:SSIV:NOST mang gen mã hóa
enzyme tăng cường sinh tổng hợp tinh bột ở củ sắn dưới sự điều khiển của
promoter đặc hiệu C54.
3. Chuyển 2 cấu trúc vector pK7WG2D-35S:SSIV:T35S và pBI121-C54:
SSIV:NOST mang gen SSIV liên quan đến trao đổi chất tinh bột ở thực vật vào cây
thuốc lá. Mức độ tích lũy tinh bột trong rễ của các dòng thuốc lá chuyển gen mang
cấu trúc pBI 121 – C54:SSIV: NOST cao hơn nhiều so với những dòng thuốc lá
chuyển gen mang cấu trúc pK7WG2D-35S:SSIV:T35S và cao hơn 27,7 - 65,7% so
với cây WT.
4. Xây dựng quy trình chuyển gen tăng cường sinh tổng hợp tinh bột SSIV
dưới sự điều khiển promoter đặc hiệu ở củ C54 vào mô sẹo phôi hóa (FEC) của
giống sắn KM140 thông qua A.tumefaciens. Kết quả thu được 7 dòng sắn giống
KM140 dương tính khi kiểm tra với PCR, tỷ lệ cây chuyển gen mang cấu trúc pBI
121 – C54:SSIV: NOST đạt 0,46%.
KIẾN NGHỊ
1. Tiếp tục phân tích, đánh giá khả năng tăng cường sinh tổng hợp tinh bột ở
củ những cây sắn chuyển gen còn lại trong điều kiện nhà lưới.
2. Ứng dụng quy trình chuyển gen vào cây sắn thông qua FEC nhờ
A.tumefaciens vào một số giống sắn khác của Việt Nam.
