Luận án Nghiên cứu sử dụng thực khuẩn thể trong phòng trị bệnh thối hạt trên lúa do vi khuẩn Burkholderia glumae
- Phân lập được 112 dòng TKT và 60 dòng vi khuẩn gây bệnh thối hạt tại chín tỉnh
đồng bằng sông Cửu Long của Việt Nam.
- Đã chọn được 8 dòng thực khuẩn thể (ФBurTV25a, ФBurDT46, ФBurDT47a,
ФBurDT47b, ФBurDT48a, ФBurVL34, ФBurVL39, ФBurAG58) có phổ kí sinh rộng
trên 75% trong tổng số vi khuẩn gây bệnh thối hạt.
- Xác định 6 dòng vi khuẩn gồm BurVL21, BurDT46, BurDT50, BurDT51,
BurKG52 và BurKG57 mẫn cảm với các dòng thực khuẩn thể phân lập khoảng trên 55%,
đồng thời xác định được 6 dòng vi khuẩn này là loài Burkholderia glumae bằng kỹ thuật
sinh học phân tử với cặp mồi đặc hiệu 1416S/1414A. Trong đó, dòng vi khuẩn BurDT46
được phân lập tại Đồng Tháp gây bệnh thối hạt cao hơn các dòng vi khuẩn còn lại trên
giống lúa OM4900 trong điều kiện nhà lưới
- Tuyển chọn được 4 dòng TKT (ФBurAG58, ФBurDT47a, ФBurDT48a,
ФBurVL34) có khả năng phân giải B. glumae DT46 gây hại cao nhất.
- Bốn dòng TKT đơn (ФBurVL 34, ФBurAG58, ФBurDT47a và ФBurDT48a) cũng
như hỗn hợp 4 dòng TKT đều cho hiệu quả giảm bệnh thối hạt trên 31% ở điều kiện nhà
lưới. Trong đó, dòng TKT ФBurAG58 cho hiệu quả giảm bệnh cao trên 70% khác biệt so với
các nghiệm thức còn lại. Ngoài ra mật số TKT ở 108 pfu/ml với thời điểm áp dụng TKT 2 giờ
trước khi lây bệnh hay phun TKT kết hợp 2 giờ trước khi lây bệnh và 5 ngày sau khi lây
bệnh cho hiệu quả giảm bệnh cao.
- Về hình thái của cả 3 dòng TKT (ФBurVL 34, ФBurAG58 và ФBurDT47a) dưới
kính hiển vi điện tử (TEM) có đầu là khối đa diện và đuôi ngắn thuộc họ Podoviridae. Kết
quả giải trình tự toàn bộ genome xác định được 3 dòng TKT đều thuộc nhóm TKT độc
mang tính an toàn khi ứng dụng phòng trị ở điều kiện ngoài đồng.
