Luận án Nghiên cứu tạo chủng vi khuẩn vibrio parahaemolyticus đột biến giảm độc lực nhằm phát triển vắc - Xin phòng bệnh hoại tử gan thận trên một số loài cá biển
Cá mú chấm cam (E. coioides) được tiêm dòng V. parahaemolyticus L4650
vào phúc mạc dưới da, với liều lượng 100 µl/con, nồng độ 105 CFU/ml. Với liều
lượng này, cá được tiêm vắc-xin có tỉ lệ sống sót là 100%. Sau khi tiêm vắc-xin,
cá mú chấm cam được tiêm công cường độc bằng chủng dại LBT6 ở 3 liều lượng
tương ứng 5LD50 (14.288 CFU/con); 10LD50 (142.889 CFU/con) và 100LD50
(1.428.890 CFU/con). Trong đó, giá trị LD50 của chủng LBT6 là 105,155 tương
ứng với nồng độ 142.889 CFU/ml; liều lượng tiêm trên mỗi con cá là 100 µl/con.
Ở 15 ngày sau khi tiêm vắc-xin, khi cá mú chấm cam được công cường độc
bằng chủng vi khuẩn dại LBT6 ở ba liều công cường độc khác nhau (5 LD50; 10
LD50; 100 LD50), tỉ lệ cá chết được ghi nhận lần lượt là 0%; 1,67% và 3,45%,
tương ứng với tỉ lệ bảo hộ (RPS) là 100%; 98,1% và 96,91% (bảng 3.15). Kết quả
công cường độc sau 2 tháng tiêm chủng cũng tương tự, tỉ lệ bảo hộ tương ứng lần
lượt là 100%, 97,03% và 96,27%. Ngược lại, đối với cá không được tiêm chủng,
khi được công cường độc bằng chủng dại gây bệnh LBT6 tỉ lệ cá chết ghi nhận
được rất cao (76,67 - 97,33%). Phân lập lại vi khuẩn từ các con cá bị chết khi
được công cường độc với LBT6 cho kết quả 97% (98/101) mẫu cá chết đều xuất
hiện các khuẩn lạc mầu xanh đậm đặc trưng chủng V. parahaemolyticus trên môi
trường TCBS.
