Luận án Nghiên cứu bệnh E. Coli trên vịt ở một số tỉnh đồng bằng sông Cửu Long: Khảo sát tình hình, đặc điểm vi khuẩn gây bệnh và thử nghiệm biện pháp phòng trị
Bệnh do vi khuẩn E. coli trên vịt tại ĐBSCL là phổ biến, bệnh nguy hiểm, với tỷ lệ bệnh và tỷ lệ chế khá cao . Triệu chứng và bệnh tích bệnh do E. coli trên vịt khá phức tạp, xuất hiện ở nhiều cơ quan khác nhau như gan (79,39%), lách, tim, ruột, túi khí, túi mật, túi lòng đỏ,. Đặc biệt, bệnh tích ở gan phổ biến nhất với tỷ lệ 79,39% và với nhiều kiểu bệnh lý khác nhau như viêm sưng, hoại tử, có màu xanh mật, nhạt màu, phủ bởi fibtrin. Bệnh E. coli gây tổn thất kinh tế khá lớn trong chăn nuôi vịt với tỷ lệ bệnh 9,09% và tỷ lệ chết 5,86% trong tổng đàn, tỷ lệ chết trong số vịt bệnh lên đến 64,44%. Tỉnh Đồng Tháp là địa phương có tổn thất do bệnh lớn nhất so với các địa phương khác với tỷ lệ bệnh do E. coli trong tổng đàn là 18,36% và tỷ lệ chết là 15,37% cao hơn nhiều so với trung bình của tất cả địa phương khảo sát Bệnh E. coli xảy ra quanh năm, nhưng số ổ dịch thấp vào những tháng cuối năm (11 và 12), bệnh xảy ra trên vịt ở mọi lứa tuổi, phổ biến nhất là ở vịt nhỏ hơn 1 tháng tuổi, nhưng hao hụt nhiều nhất là ở lứa tuổi từ 31-60 ngày tuổi. Hình thức chăn nuôi chạy đồng và vịt nuôi chuyên trứng có tỷ lệ bệnh và chết cao hơn so với vịt nuôi nhốt và vịt nuôi theo hướng thịt. Vịt nuôi với qui mô đàn quá nhỏ (50-200 con) hoặc quá lớn (>5000) có tỷ lệ và chết cao hơn so với qui mô vừa. Nhóm huyết thanh E. coli phổ biến nhất trên vịt ở ĐBSCL là O2 và O78. Vi khuẩn E. coli phân lập trên vịt tại vùng ĐBSCL có khả năng đề kháng cao với một số loại kháng sinh phổ biến như streptomycin (65,86%) và ampicillin (63,88%). Nhưng nhạy cảm cao với colistin (90,29%) và amikacin (73,75%). Nghiên cứu cũng phát hiện sự hiện diện của 5 gene đề kháng kháng sinh (TEM, SHV, TetA, Sul1, aadA1) tỷ lệ cao nhất là TetA (73,33%) và thấp nhất là SHV (22,67) và 5 gene mã hóa yếu tố độc lực (FimH, ColV, VAT, Iss, HlyA) trong các mẫu phân lập, phổ biến nhất là FimH (44,4%). Trình tự nucleotide của các gene mã hóa yếu tố đề kháng kháng sinh hoàn toàn tương đồng với nhau và tương đồng rất cao với các gene tham chiếu trên GenBank. Tiêm bắp amikacin với liều 10 mg/kg P, liên tục trong 5 ngày có hiệu quả tốt trong phòng và trị bệnh do E. coli trên vịt.

Xác định liều gây chết 50% (LD50) trên vịt
LD50 được tính toán dựa theo công thức của Reed and Muench (1938):

Trong đó: PD (proportional distance) là khoảng cân đối
LD50 là liều gây chết 50% động vật thí nghiệm.
Kết quả xác định liều gây chết LD50 trong nghiên cứu này như sau:

Sau khi thí nghiệm kết quả đã được tính: LD50 = 107,64 CFU/ml.
Chuẩn bị vi khuẩn
Vi khuẩn E. coli dùng trong thí nghiệm là nhóm huyết thanh APEC (O78) phân
lập trên vịt bệnh trong nội dung 1 và đã được xác định sự hiện diện của 5 gene mã hóa
yếu tố đề kháng kháng sinh và 5 gene mã hóa yếu tố độc lực trong nội dung 2. Đồng
thời, nhóm huyết thanh APEC (O78) dùng trong thí nghiệm đã được xác định liều
LD50 (107,64 CFU/ml). Vi khuẩn E. coli thuần được cấy chuyển sang môi trường NA
nuôi cấy trong 24 giờ ở 37oC. Lấy khuẩn lạc nuôi tăng sinh trên môi trường NA ở
37oC trong vòng 24 giờ. Dùng que cấy vô trùng lấy tất cả khuẩn lạc cho vào ống
nghiệm có chứa 10 ml dung dịch PBS lắc đều bằng máy vortex. Sau đó, tiến hành pha
loãng để đạt được nồng độ cần cho thí nghiệm. Xác định mật độ vi khuẩn bằng cách so
sánh độ đục của canh trùng trong ống nghiệm với độ đục của ống dung dịch chuẩn
0,50 McFarland (tương đương 1 x 107,64 CFU/ml).
Độ đục chuẩn 0,50 McFaland được pha theo tiêu chuẩn McFarland (McFarland
standards) (Trần Linh Thước, 2009).
Dung dịch BaCl2 1% 0,5 ml
Dung dịch H2SO4 1% 99,5 ml
